Good definitions by Leah and Rusty.
To take Rusty's definition of tone even further. I could pick up Derek's guitar as it was set for a concert and play a passage of an ABB tune/exact same notes as him (obviously he would play it better, but assuming I played it with the same ability), it would still sound a bit different when Derek played even though we are using the exact same rig. This is because of the difference in our fingers, the way he grabs the notes, the way he picks the notes, and so on...to me this is also a factor in the tone of a guitar player, it is not just the sound of the guitar, amp, effects, use of volume, and so on. You can hear the sound of the guitar and amp when the roadie strums it during the soundcheck, but you won't really hear the tone till the player plays it.
For example, Jimmy Herring has killer tone. Someone mention Mark Knopfler yesterday, great example of unmistakable tone.
